Double immunofluorescence staining, with antibodies against the mannose receptor and vascular endothelial growth factor receptor (VEGFR-3), a known marker of lymphatic endothelium, shows the word presence of the mannose receptor on lymphatic endothelium. In this merged image, colocalization of the mannose receptor and VEGFR-3 is seen as a yellow signal. The presence of the mannose receptor on lymphatic endothelium mediates binding of lymphocytes. The binding requires L-selectin, which functions as a ligand for the mannose receptor. Interaction between L-selectin and mannose receptor is the first molecularly defined mechanism mediating lymphocyte binding to lymphatic endothelium.
